Sodium Benzoate

Sodium Benzoate Capsule

 

 

English product name: Pure Sodium Benzoate Capsules R Chinese product name: Pure Sodium Benzoate Capsules Main ingredient: sodium benzoate Dosage form: capsule 250mg.

 

Indications: Adjuvant treatment of congenital nonketotic hyperglycinemia.

 

Pharmacological mechanism: Sodium Benzoate can reduce blood ammonia concentration in patients with congenital urea cycle abnormalities. Its mechanism of action includes the conjugation reaction of amino acid sylation. First, hippurate is formed by the conjugation of benzoate and glycine.

 

When hippurate is combined with nitrogen, these compounds are then excreted in the urine, thereby reducing nitrogen accumulation and lowering blood ammonia levels.

 

The synthesis of Hippurate requires two steps, ATP and coenzyme A are required to participate in the formation of an intermediate product of acyl-coenzyme A and then undergo transacylation with glycine.

 

Treating patients with nonketotic hyperglycinemia with high doses of benzoate can reduce the amount of glycine in blood and brain, improve epilepsy, and then improve the quality of life of patients alive.

 

Pharmacokinetics: 1. This product does not conduct pharmacokinetic studies on patients such as neonates, infants, and children, but only conducts pharmacokinetic studies on normal adults. The results show that within 1 hour after oral administration of sodium benzoate, the blood The concentration of benzoate reaches the highest peak.

 

2. Within 24 hours, about 80-100% is excreted by the kidneys through hippurate conjugation, so the main place of metabolism is the kidneys.

 

Contraindications: So far there are no reports of relevant contraindications.

 

Side effects: gastrointestinal discomfort, similar side effects to salicylic acid.

 

Pregnancy Grading: Data not yet established.

 

Interaction: 1. Penicillin: It will reduce the effect of sodium benzoate.

 

2. Valproic acid: It can cause hyperammonemia and reduce the therapeutic effect of sodium benzoate.

 

3. Probenecid is known to inhibit the transmission of many compounds in the renal tubules and affect the excretion of sodium benzoate conjugates in the kidney.

 

Notes: 1. According to in vitro tests, benzoate will compete with bilirubin to combine with albumin, so it should be used with caution in patients with neonatal bilirubinemia.

 

2. Patients with congestive heart disease should use it with caution.

 

3. Since sodium benzoate itself contains sodium ions, patients with hypernatremia should use it with caution.

 

4. Patients who are allergic to sodium benzoate should avoid using this medicine.

 

Dosage;

 

1. The starting dose is 250mg/kg/day, taking 3~6 times on average. In addition, depending on the degree of blood glycine decline, the dose can be increased to 750mg/kg/day, and the total daily dose should not exceed 10g.

 

2. When taking sodium benzoate, it can be combined with a low-glycine diet and other drugs to achieve better results.

 

Storage: Store in a tightly covered container below 25°C and avoid direct sunlight.
